Control of Erythropoiesis in Rats With Adjuvant.induced

چکیده
In order to characterize the defect in erythroid homeostasis in chronic inflammatory states, the relationship between erythropoietin production and erythropoietic response was examined in rats with adjuvant disease. Exposure of adjuvant-injected rats to graded levels of lowered barometric pressure induced increases in plasma erythropoietin which were significantly less than those measured in normal animals similarly stimulated. Erythropoietin inhibitors were not detected by in vitro or in vivo assay techniques: the biological activity of ovine erythropoietin was not modified by incubation with plasma from rats with adjuvant disease; the erythropoietic response of ex-hypoxic polycythemic mice to erythropoietin was not compromised by injections of test plasma; and the burst of erythropoiesis induced in exhypoxic polycythemic mice by a hypobaric stimulus was not modified by plasma given prior to or at various intervals after hypobaric exposure. Exogenous erythropoietin elicited nearly identical increases of radioiron incorporation in normal and adjuvantinjected rats whose endogenous erythropoietin was suppressed by hypertransfusion. It is concluded that the diminished erythropoietic response to anemia in adjuvant-induced chronic inflammation results from a relative failure in the production of biologically active erythropoietin.

Serum erythropoietin (EPO) concentration was measured by an enzyme-linked immunosorbent assay (ELISA) in 31 cases of chronic renal failure (CRF), including 20 hemodialysis (HD) patients, and compared with that of 31 healthy normal controls. Each patient served as a self-control comparing his or her pretreatment level with post-treatment values. There was a significant negative correlation ...
متن کاملEffects of Recombinant Human Erythropoietin (rHuEpo) on the Blood Cells and Hematopoiesis in Spleen of Rats
Recombinant human erythropoietin is a glycoprotein hormone that stimulates erythropoiesis. In clonogenic assays of hematopoietic progenitors, high concen-trations of erythropoietin (Epo) increase CFU-E and diminish the number of granulocytes formed per culture plate. Fetal progenitors are more sensitive to these effects of Epo than progenitors from adults. We administered doses 50, 100, 200 IU/...

متن کاملEffect of testosterone propionate on erythropoiesis after experimental orchiectomy.
INTRODUCTION Androgen deficiency anemia occurs most frequently in pharmacogenic suppression of androgen synthesis or with advancing age in men. Bilateral orchiectomy is a surgical modality used in the treatment of metastatic prostate carcinoma. It is accompanied by marked decrease in circulating serum levels of androgens. AIM The aim of the experimental study was to determine the effect of su...

Erythropoiesis in spleens of lethally irradiated Lewis rats grafted with 4-35 X 10(6) syngeneic marrow cells was inhibited or delayed during the test period of 5 days; this was in marked contrast to observation in irradiated mice. The mechanism of this inhibition was the subject of this study. Pretreatment of recipients 9 days prior to irradiation with the cytotoxic drugs cyclophosphamide (CY),...
